Next-generation ADCs, built on reversible, site-selective chemistry.

OPAL is a bioconjugation platform for building better-defined ADCs. It controls exactly where and how payloads attach and can take them off again, making conjugation and purification a single step.

The technology

Stable bioconjugation that goes both ways

OPAL is a proprietary bioconjugation technology that installs payloads at defined sites on an antibody scaffold. By controlling exactly where and how many drug molecules are conjugated, OPAL eliminates the heterogeneous mixture of drug-to-antibody ratios (DAR) that limits conventional ADCs.

The stable bond is also reversible on demand. The same chemistry that installs a payload can release it, so conjugation and purification run as one operation rather than two.

The result is a well-defined, homogeneous product with predictable pharmacokinetics, improved developability, and a clearer path from discovery to clinical candidate.

The platform

One chemistry, many capabilities

OPAL conjugation gives medicinal chemistry teams precise control over where, how much, and what is attached.

Site-selective

Conjugation is directed to defined sites on the antibody for truly homogeneous products.

Reversible

The ligation runs in reverse on demand, making the chemistry uniquely useful for in tandem purification and conjugation.

Controlled DAR

Set and reliably reproduce the exact drug-to-antibody ratio your therapeutic requires.

Impact

Why it matters for ADC development

Conjugation heterogeneity is one of the biggest sources of unpredictability in ADC manufacturing. OPAL turns that variability into a controlled parameter.

Fewer unit operations

Conjugation and purification share one chemistry, removing a separation step from the downstream process.

Smoother scale-up

Low batch-to-batch variability supports robust process development and CMC timelines.

Predictable PK

Controlled payload loading leads to more consistent pharmacokinetic profiles across batches.

Building better ADCs

We believe streamlined, site-selective conjugation should be the foundation of every ADC, yielding conjugates with defined DAR by design, homogeneous by default, and reproducible at any scale.
